Library prep

Batching

Batching is the practice of grouping multiple samples so they can be processed, amplified, or sequenced together in a single run.

What is batching in NGS?

Traditionally a pre-amplification step, since samples had to be grouped by expected input to share one cycling program. Per-well control removes that constraint, so batching is not necessary when using AutoNorm.

Read Balancing

Read balancing is the process of adjusting library ratios so every sample receives a comparable share of the sequencing output.

Quantification

Quantification is the process of measuring the concentration of DNA in a sample, typically before pooling and sequencing.

Pooling

Pooling is the process of combining multiple indexed libraries into one tube so they can be sequenced together in a single run.

Normalization

Normalization is the process of equalizing the concentration of different DNA libraries so each is represented evenly when pooled.

Library Preparation

Library preparation is the multi-step process of converting DNA or RNA samples into a collection of sequencing-ready fragments.

Library Clean Up

Library clean up is the step that removes leftover reagents and unwanted reaction byproducts from a library before sequencing.
