PCR artifacts

Plateau

Plateau is the late stage of PCR where reaction components are exhausted, amplification stalls, and artifacts begin to form.

What is the plateau phase in PCR?

Amplification curves flatten here, so extra cycles add artifacts rather than yield. Stopping before plateau is the single most effective way to keep a library clean.

Size Bias

Size bias is the tendency of PCR to preferentially amplify shorter fragments, distorting the original fragment size distribution.

Recombination

Recombination is the unintended shuffling of genetic material between fragments, often driven by over-amplification.

Primer Dimers

Primer dimers are artifacts formed when two primer molecules hybridize to each other instead of to the target DNA template.
